Construction of a Fear Scoring System in Laying Hens and Its Association with Immune Status and Gut Microbiota

Compared to the tonic immobility test (TI), commonly used to assess fear status, the open field test (OFT) provides a more comprehensive evaluation of fear levels in hens. Based on key fear-related indicators from previous studies, we developed a systematic fear scoring system and explored its association with immunity and gut microbiota. Ninety-six individually caged Lindian-hens (60 weeks old) underwent an OFT at 62 weeks and a TI at 64 weeks. Fear scores correlated positively with TI duration and negatively with vocalization and exploration, validating the system.

Hens from different fear score groups (1, 3, 5 points, n=6/group) were selected for serum analysis. The 5-point group compared with the 1-point showed significantly higher IL-1β, IL-6, TNF-α, and IgY, but lower IL-10 and IL-2(P < 0.05). 16S-rRNA sequencing revealed lower bacterial richness in the 5-point group, dominated by Firmicutes, while the 1-point group was enriched in Bacteroidetes.

Microbial α-diversity in the 1-point group was higher than 5-point while β-diversity revealed distinct microbial structures between the 1- and 5-point,with the 3-point group being intermediate. Gut immune analysis indicated increased IL-1β, IL-6, IFN-γ, and IgY mRNA in the 5-point group, whereas IL-4, IL-8, IL-10, and IgA were reduced(P < 0.05). Correlation analysis linked Bacteroides to IL-10 and IgA (positive) and IL-1β and IgY (negative), while Lactobacillus and Prevotella correlated with IL-6 and TNF-α, respectively(P < 0.05, |r| > 0.6).

This study establishes a fear scoring system and highlights interactions between fear, immunity, and gut microbiota, offering insights into poultry welfare.
