The resource utilization of livestock and poultry manure are critical for sustainable agricultural development. Anaerobic digestion is one of most successful technologies for livestock and poultry manure valorization. However, the commercial application of biogas produced from anaerobic digestion faces several challenges, such as limited methane content (<60%), extensive purification energy consumption, and limited grid connection. In recent years, microbial electrochemical systems (MESs) have been extensively researched and developed, offering a promising method for biogas upgrading. However, successful application of MESs to biogas upgrading requires a deeper understand on their potential mechanism and optimization of operational performance. To this end, the mechanism of biogas upgrading by MESs, involving MESs fundamentals, extracellular electron transfer and conversion pathways from CO2 to CH4 via functional enzymes of microorganisms, were analyzed and discussed. In particular, quantitative analysis of electron transfer distribution in different pathways has been summarized and emphasized. At last, current challenges and prospects for further mechanism inquiry, optimization of upgrading process and practical application were summarized.
