Study on Particulate matter and Microbial community changes in multi-layered caged broiler house in summer

The aim of this study was to investigate the distribution characteristics of particulate matter (PM) and microbial community in multi-layer caged broiler house in summer. In this experiment, the PM(TSP, PM10, PM2.5 and PM1) in the broiler house at day 1, 21 and 49 were collected and measured. The microbial DNA in TSP was extracted, and the V3-V4 region of bacterial 16S rDNA and the ITS2 region of fungal were amplified by PCR and sequenced, respectively. The results revealed that the concentrations of TSP, PM10, PM2.5 and PM1 inside the house were significantly higher than those outside the house at day 1, 21 and 49. The concentrations of TSP, PM10, PM2.5 and PM1 indoors increased significantly from 0.092 mg/m3, 0.063 mg/m3, 0.053 mg/m3 and 0.053 mg/m3 on day 1 to 0.998 mg/m3, 0.541 mg/m3, 0.426 mg/m3 and 0.423 mg/m3 on day 49 (P<0.05). On day 21 and 49, the concentrations of all PM in the house increased significantly along the front, middle and back area. The changes of bacterial and fungal community composition in the house were different. The Chao1 index of bacteria significantly increased, while that of fungi significantly decreased as the ages of broiler increased. The main bacterial phyla were Bacillota, Pseudomonadota, Bacteroidota and Actinomycetota. The relative abundance of Bacillus significantly increased from day 1 to day 49 while the other three phyla significantly decreased. Ascomycota and Basidiomycota were the two main dominant phyla of fungi, and Ascomycota was the preponderant phylum through the broiler growth stages.
