This paper takes Wuhan as the research area, with collecting NPP-VIIRS night light remote sensing data and POI data of Wuhan in 2018 as the data source, and utilizing the overlay analysis and two-factor analysis to visualizing the relationship between spatial coupling relationship and urban spatial structure and identifying the areas of similarities and differences in coupling relations. Based on this, the development of the urban structure of main axis in Wuhan Yangtze River is discussed. The main conclusions are drowned as following: (1)Under the macroscopic scale of the city, the overall trend of nighttime light data is in accordance with the POI data, and highly consistent coupling relationship between nighttime light data and POI data is revealed, and the area with the same spatial coupling relationship accounts for 87.7%, which well characterized the overall planning of Wuhan City; (2)Different coupling relation areas are mainly in urban fringe, new city center, urban riverside, urban ecological protection areas, scenic spots and other areas; (3) As an essential riverside city, the close relationship between the urban spatial structure and the core section of the main axis of the Yangtze river has been revealed, while the relationship of the extended section of the main axis is not obvious.
