Programmed cell death (PCD) is one of the most important cell process, plays crucial role in morphogenesis, defense responses to pathogens and other stress conditions Previous studies have demonstrated that S1/P1 nucleases are mainly responsible of nucleic acids degradation in plants. But to this date only one degrading nuclease expression pattern was studied in A.thaliana – BFN1 from S1-like family proteins. We examined rest of S1 nucleases family members using GUS reporter gene system.
Our results suggest very high diversity of expression patterns of this family both in different time and tissues. Promoters of members of S1-like family show distinctive but in some cases overlapping expression pattern.
There is also one more degrading nucleases family from A.thaliana - Staphylococcal-like (SNc-like) nucleases. They are neutral calcium-dependent enzymes with cell membrane localization. Although their function remained unknown. To find out their potential role we performed GUS reporter assay for both two members of this gene family.
Our results show that both SNc-like nucleases promoters show high expression during leaf senescence (overlapping with ENDO1, ENDO3 and ENDO5 promoter), but only CAN1 is expressed in all tissues undergoing PCD.
It's suggesting that role of degrading nucleases in plants is more complex that it was previous assumed.
